چکیده

decreased to 2276±288 nm at the end of 120 min having a polydispersity over 0.7 for both of the incubations. Among the CLDC10 and CLDC11 prepared at the highest ratios (1:0.186 w/w) the complexation process was the fastest and the size observed between 2900 nm to 3044 nm remained nearly unchanged with CLDC10, whereas a progressive increase and then decrease was observed with CLDC11. The measured size of CLDC10 was 2412±62.7 nm at the end of 120 min, however the polydispersity index was over 0.7. The complexation was completed at all incubation periods for both complexes as confirmed by the gel electrophoresis analysis.

Effect of Helper Lipids on Stability and Transfection Activity of Lyophilized Lipoplex Formulations of Antisense and DOTAP Nanoliposomes

Survivin, an inhibitor of apoptosis protein is highly expressed in most cancers and considered as an attractive target for cancer antisense therapy. To vectorize antisense molecules, cationic nanoliposomes are generally used; however, their complexes are too instable, during shelf-life and upon exposure to blood components and extracellular matrix, to be used in-vivo. The present study a...

Modeling of cationic lipid-DNA complexes.

Cationic lipid-DNA complexes, often referred to as lipoplexes, are formed spontaneously in aqueous solutions upon mixing DNA and liposomes composed of cationic and nonionic lipids. Understanding the mechanisms underlying lipoplex formation, structure and phase behavior is crucial for their further development and design as non-viral transfection vectors in gene therapy. From a physical point of...

Physico-Chemical Characteristics of Lipoplexes Influence Cell Uptake Mechanisms and Transfection Efficacy

BACKGROUND Formulation of DNA/cationic lipid complexes (lipoplexes) designed for nucleic acid delivery mostly results in positively charged particles which are thought to enter cells by endocytosis. We recently developed a lipoplex formulation called Neutraplex that allows preparation of both cationic and anionic stable complexes with similar lipid content and ultrastructure. METHODOLOGY/PRIN...

Trends in lipoplex physical properties dependent on cationic lipid structure, vehicle and complexation procedure do not correlate with biological activity.

Using a group of structurally related cytofectins, the effects of different vehicle constituents and mixing techniques on the physical properties and biological activity of lipoplexes were systematically examined. Physical properties were examined using a combination of dye accessibility assays, centrifugation, gel electrophoresis and dynamic light scattering. Biological activity was examined u...

Synergy between cationic lipid and co-lipid determines the macroscopic structure and transfection activity of lipoplexes.

The large number of cytofectin and co-lipid combinations currently used for lipoplex-mediated gene delivery reflects the fact that the optimal cytofectin/co-lipid combination varies with the application. The effects of structural changes in both cytofectin and co-lipid were systematically examined to identify structure-activity relationships. Specifically, alkyl chain length, degree of unsatura...
